科研成果
在基于納米技術新型藥物遞釋系統的應用基礎研究及將基礎研究成果向臨床應用轉化方面取得了一系列突出成績,主要包括:
① 探索了顺序靶向策略,将药物-靶向多肽键合物包载到可穿过血脑屏障的转铁蛋白修饰胶束中,该设计在实现跨血脑屏障转运后,可通过RGD 识别肿瘤细胞上高表达的整合素受体,实现脑胶质瘤靶向,显著提高了脑胶质瘤治疗效果,降低了毒副作用。
② 实现了穿膜肽自组装,阐明了穿膜多肽纳米载体在跨膜递送方面的独特优势;揭示了在药物-穿膜肽键合物设计中,药物连接位点、疏水性、连接密度、亲水序列对其理化特性和生物效果的影响,阐明了药物多肽键合物设计要点,这些研究为两亲性多肽的理性设计奠定了扎实的基础。
③ 设计构建了包载洛沙坦的两亲性多肽,并使其形成水凝胶,探索了两亲性多肽水凝胶在肿瘤微环境调节,减少肿瘤相关成纤维细胞的数量和活性,增加纳米载药系统向肿瘤的分布和渗透方面的作用,证实该多肽水凝胶能够与纳米载药系统协同增效。
④ 设计构建了光刺激响应性功能纳米粒,利用光照产生的光动力治疗特性,杀死肿瘤浅表快速生长的细胞,利用光动力促进药物溶酶体逃逸特性,增加化疗药物和siRNA 的胞浆释放和疗效,克服肿瘤异质性;设计构建了诊疗一体的仿细胞毒性纳米粒,能够实时检测纳米载药系统在肿瘤部位蓄积和激活,通过联合光动力和化疗,有效提高了转移性乳腺癌治疗效果。
    近年来在国际重要学术期刊(IF>3.0)上发表SCI論文20余篇,其中IF>10的論文9 篇,IF=5-10间的論文8篇。以第一作者或通讯作者身份发表的代表性論文包括: Chem Soc Rev(1篇,IF=40.18),Adv Mater(1篇,IF=21.95),ACS Nano(2 篇,IF=13.70), Biomaterials(1 篇,IF=8.80),Theranostics(3 篇,IF=8.53),Curr Opin Biotechnol(1篇,IF=8.38),J Control Release(1 篇,IF=7.87),Chem Commun(1 篇,IF=6.29);論文累计他引1200 余次。
